The paper that first identified specific receptor targets for peat-derived substances. Published in Phytomedicine, it is one of the most consequential pharmacology studies in the peat literature — the point where peat went from “natural agent with unclear mechanism” to “agent with known receptor pharmacology.” Referenced by nearly every subsequent peat pharmacology paper, including Beer 2003b, which extended this work to human skin permeation.

What Was Studied

The researchers separated water-soluble fractions from peat extract — specifically fulvic acids and ulmic acids — by pH and solvent. These fractions were then tested for their effect on spontaneous contractile activity (SCA) in guinea pig stomach smooth muscle fibers, a standard bioassay for receptor pharmacology. By testing separated fractions rather than whole peat extract, the study could attribute effects to specific compound classes.

The Receptor Findings

Fulvic and ulmic acid fractions from peat act as partial agonists on two receptor types:

  • α2-adrenergic receptors — involved in smooth muscle tone, vascular regulation, and pain signalling
  • D2 dopamine receptors — involved in smooth muscle relaxation and vasodilation

Different fractions showed different receptor profiles and effects on contractile activity — consistent with Beer 2003b’s later finding that only specific HPLC fractions of peat extract permeate skin and exert pharmacological effects.

Why This Matters

Before this paper, the evidence for peat’s biological activity was observational: patients improved. Beer 2000 established the mechanistic link — peat contains compounds that bind to specific receptors and trigger measurable physiological responses. This moved the field from empirical observation to receptor pharmacology, providing the molecular rationale for the anti-inflammatory, analgesic, and muscle-relaxing effects reported in clinical trials.
